Barebones Guide to the HP UV-VIS Spectrometer:
Spectrum Scanning Mode

  1. Turn on the UV and the computer. Wait about 10 minutes for the lamp to warm up.

  2. In the software, select “General Scanning”.

  3. To change the wavelength range, hit the F4 button. The instrument can scan from 190-820 nm.

  4. Insert your blank (solvent) cuvette in the sample holder and press the F2 button to measure your blank. The blank will automatically be subtracted from the sample scans. NOTE: You may want to re-acquire a blank every 2-3 samples in order to be sure the background is not drifting.

  5. Insert your sample cuvette into the sample holder and press the F1 key to acquire your sample.

  6. To label peaks, press the F2 key.

    1. To automatically label your peaks, press the F2 button.

    2. You may need to manually select your peaks if your sample is very dilute and the “Peak Find” option above picks a lot of noise. To do this, move the cursor on your spectrum using the left of right arrow button. Press F1 when you are at the maximum absorbance of your peak of interest.

  7. To save your file, press F10 until you are at the main screen. Press F6, and select “Store Data File”. Enter a file name (no more than 8 characters!) at the prompt.

  8. To overlay your spectra (if you have more than one) you can press the F5 key, then select “Overlay Spectrum” and press Enter.

  9. To return to your sample spectrum, press F9.

  10. When you are finished, press F10 to exit the software, and hit Enter. At the software bootup screen select “Exit” and hit enter.

  11. To transfer your files to a floppy disk, insert a 3.5” floppy into the drive.

    1. Type “cd \HPUV\Data” at the DOS prompt.

    2. Type “copy filename.wav a:”, where filename is your filename.

    3. Repeat steps a and b as needed.

  12. When you are finished, you may turn off the instrument and computer by pressing the power button on each.
